Category:Lava Records singlesThe goal of this project is to develop a three-dimensional (3D) in vitro tissue culture system that will model intact tissue of the fetal rabbit developing lung. This will enable us to explore cell-cell and cell-matrix interactions in vitro in the context of developing tissues. The model will be used to determine the relative contribution of epithelial cells and mesenchymal cells to the production of fetal lung surfactant. This project will also determine whether surfactant can be synthesized by rabbit lung epithelial cells in culture. The model will be used to study changes in cell-cell and cell-matrix interactions in developing fetal lung, as well as to study the influence of the microenvironment on the production of surfactant by lung cells. One of the major barriers to progress in the study of the lung is the lack of a good model system. Although organ culture has been widely used in studies of the fetal and neonatal lung, it has not been widely used in studies of the fetus and newborn lung because of the poor survival and maturation of the epithelial cells. Our work with organ culture shows that rabbit fetal lung epithelial cells can survive in vitro, but that they do not produce surfactant or develop a polarized epithelial layer.
